The University Instrumentation Center's shared light microscopy facility is located in Rudman Hall 157. Any researchers are encouraged to make use of this facility, which includes both upright and inverted fluorescence microscopes. Training and access to the facility can be provided by the UIC upon request.
Olympus IX81 Inverted Fluorescence/Brightfield Microscope
The Olympus IX81 inverted fluorescense/brightfield microscope has a motorized stage and stage inserts to accommodate slides or multiwell plates. It has six objective lenses: 4X/0.13, 10X/0.40, 20X/0.70, 40X/0.85, 60X/0.90, and 100X/1.40. The 100X is an oil-immersion lens and the 60X can be used with differential interference contrast (DIC) optics. For fluorescence imaging, it has three filter sets suitabile with DAPI-, GFP-, and CY3-like fluors. Images are captured with an Olympus DP80 combined monochrome/color camera, with fluorescent images usually taken using the more sensitive monochrome chip, then artificially colored, and transmitted light images taken with either color or monochrome chips. Image capture and other aspects of microscope operation are controlled with Olympus cellSens Dimension software with added Well Navigator and Multi Position software modules to facilitate and automate image acquisition from multiwell plates.
Zeiss Axioplan 2 Imaging Upright Fluorescence/Brightfield Microscope
The Zeiss Axioplan 2 Imaging upright fluorescence/brightfield microscope has six objective lenses: 2.5X/0.075, 10X/0.30, 20X/0.50, 40X/1.30, 63X/1.25, 100X/1.40. The 40X, 63X, and 100X are oil-immersion lens and the 10X, 20X, 63X, and 100X can be used with differential interference contrast (DIC) optics. For fluorescence imaging, it has seven filter sets suitable with DAPI-, GFP-, Texas Red-, Rhodamine-, CY5-, QDot Green-, and QDot Yellow-like fluors. Images are captured with Zeiss AxioCam MRc5 color or Zeiss AxioCam 503 monochrome cameras. Image capture and other aspects of microscope operation are controlled with Zeiss AxioVision Rel. 4.9.1 software.
Olympus SZX9 Stereomicroscope
The Olympus SZX9 Stereomicroscope is?especially useful for inspecting/imaging 3D macroscopic objects; 1x objective lens, 6.3-57x magnification; illumination by reflected light (fiber optics) from above sample and/or by transmitted light from below (brightfield or darkfield); images captured with Olympus Q-Color 3 digital camera and Q Capture Pro 51 software.
